What are some good games that provide a gameplay experience you can't get anywhere else?

What era are we talking about here? Chu Chu Rocket for DC is pretty unique, and extremely fun if you have 3 friends around.

For one of a kind games I look to the computer classics. Namely C64... Impossible Mission, Archon, Skool Daze, Frankie Goes to Hollywood, Project Firestart, Forbidden Forest 1 & 2, Tusker... Not to mention all the classic Lucasfilm games... And those are only the ones that come to the top of my head...

ZX Spectrum "How to be a Complete Bastard". I can't say I can think of another game where the aim is to ruin a house party for everyone.

And then there's "Mad Nurse" for the c64... A game where you can electrocute, drown, and use stun gas on new born babies...

Alpha Waves for the PC, Amiga, and Atari ST. The gameplay is very different from your average platformer in that it's a sort of 3D Crush roller crossed with Super Pac Man and Mario 64. You're this robot thing who bounces on platforms and tries to find all the rooms in the particular area it's in. You also have to get keys to unlock new areas. There's also a time attack mode where you're supposed to find all the rooms before the time runs out.
